Most Antivirus softwares (If not all) are locally installed (On your PC) and access online database for virus signatures periodically (Day/Week etc).. In my opinion, the best 2 are NOD32 %26 Kaspersky..

One of the easiest ways to detect a virus is to check the registry, and the autoexec.bat, config.sys, and win.ini files. Antivirus programs check all of these places for traces of virii, and some check the Open with extensions, then the program checks file by file with an algorithm that detects variants of known virii. Some might use CRCs and checksums to simplify the process, but this just adds to the possibility for error.
